Is the biopsy diagnosis of prostate cancer an effective tool?

Has anyone ever had one? My husband is having a Thursday because the dr. found a small tumor of the prostate. The PSA test became negative, but doc still wants to do a biopsy to see if it is cancer. Is the procedure painful? Thank you in advance!
